I sperm check my males whenever they are used for a breeding.
I'm not a vet but sperm are pretty easy to see with a basic microscope.
Here's what I do...
When the male is finished breeding the female she will usually stay cushed for a few minutes. During that time I take a craft stick and scoop up some of the sticky stuff from her vulva. It's often blood-tinged but whitish looking. Put it on a microscope slide immediately and quickly look at it under the microscope. Protect your sample from the sun on your way there.
Once it's under the scope I can usually see movement at 10X magnification and clearly see individual sperm at 40X.
It's not a very scientific sperm count but I can tell if that male probably has none, some or plenty. This time of year with the heat here in the south many of the males have very low sperm counts.
